The gamma dose required to achieve SAL 10\u207b\u2076 (the standard sterility assurance level) for medical devices is typically 25 kGy as a minimum dose, with 50 kGy used for devices requiring additional dose assurance or for products with high bioburden. At these doses, every organic material in the device \u2014 including the epoxy adhesive \u2014 absorbs radiation energy that generates chemical change. Understanding those changes and specifying accordingly prevents the failure mode of a device that passes all pre-sterilization testing but degrades after the sterilization step it must survive before reaching the patient.<\/p>\n<h3>The Physics of Radiation Damage in Epoxy<\/h3>\n<p>Gamma photons interact with the polymer matrix through Compton scattering and photoelectric absorption, generating high-energy secondary electrons that deposit their energy along short paths within the material. For transparent or optically clear adhesives used in diagnostic optical assemblies, gamma-induced discoloration may be functionally unacceptable. Formulations with reduced aromatic content or antioxidant additives show less discoloration.<\/p>\n<p>Surface property changes: The adhesive surface may become slightly more brittle, rougher, or tacky after gamma, depending on the degree of oxidative degradation at the exposed surface. Characterizing adhesive properties at doses above the intended sterilization dose (for example, at 2\u00d7 the intended dose) provides evidence of robustness against accidental over-exposure.<\/p>\n<p>Dose rate effects are generally small for epoxy adhesives at the dose rates used in commercial gamma sterilization (typically 1 to 10 Gy\/second). Accelerated testing at very high dose rates may not accurately represent property changes at the low dose rates of commercial sterilization, but dose-rate effects are typically not significant enough to affect specification decisions for epoxy materials.<\/p>\n<h3>Post-Gamma Biocompatibility<\/h3>\n<p>A material that is biocompatible before gamma irradiation may or may not remain biocompatible after gamma, because irradiation can generate new chemical species \u2014 oxidation products, chain fragments, and recombination byproducts \u2014 that were not present before irradiation.<\/p>\n<p>For complete biocompatibility coverage in a gamma-sterilized device, the biocompatibility testing specimens should be irradiated at the maximum intended sterilization dose before testing. ISO 10993-12 specifies that sample preparation for biocompatibility testing should simulate the end-use condition of the material, which for a gamma-sterilized device means post-gamma specimens.
